One of the early problems of previous generation 5-HT4 agents was that it caused cardiac problems. This has made the FDA very cautious in interpreting the data. Moreover, there has been a very long lag time between the prucalopride design and clinical trial, leading many to suspect that there have been internal problems with the medication concerning side effects.
